Introduction
The reverse dot blot technique provides a foundation for highly specific probe hybridization to detect multiple genotypes or mutations at the same time; using allele-specific probes that are immobilized on the nitrocellulose membrane instead of directly on the analyzed DNA sample. This technique is used by researchers to detect many distinct sequences at the same time with high specificity and accuracy such as detecting HPV genotypes, identifying IS6110 insertion sites in the viral genome of Mycobacterium tuberculosis, and detecting α-thalassaemia mutations, lactic acid bacteria in fermented foods or intestinal infection bacteria, etc. According to statistics, there are about 200 HPV genotypes and are classified into 3 groups: high risk, likely high risk, and low risk. Genotype detection is extremely important in the screening and treatment of positive patients at high risk of cancer.
